8-Methoxypsoralen (8-MOP) is used for photochemotherapy of psoriasis and vitiligo. In therapy, called PUVA therapy, the affected skin of a patient who has previously been prescribed 8-MOP is irradiated with UV-A (> 320 nm). As a result of the photochemical reaction, 8-MOP forms a covalent bond with the pyrimidine bases of DNA and forms both monofunctional and bifunctional adducts. The latter cause 8-MOP-mediated cytotoxicity, leading to the death of psoriatic cells. However, in surviving cells, these same adducts can cause 8-MOP genotoxicity. For a comparative study of the cytotoxicity and genotoxicity of 8-MOP, the E. coli MG1655 lux biosensor (pColD-lux), carrying a recombinant plasmid with a lux operon controlled by the colD gene promoter, was first used. The colD gene (cda) is part of the E. coli SOS-regulon, which provides DNA repair and cell resistance to DNA damage. The death of bacteria was taken into account as an indicator of the cytotoxicity of 8-MOP+UV-A, and the death of bacteria was taken into account by the change in the luminescence intensity of the biosensor. Induction of 8-MOP-mediated SOS-response in bacteria depends on the dose of UV-A, and on the concentration of 8-MOP. At high UV-A doses, a 25-fold decrease in the survival of bacterial cells was detected (from 2 × 108 to 8 × 106 CFU), and in viable cells, an increase in the intensity of the SOS response by 675 times in terms of 106 cells. Thus, it was shown that in the bacterial test system, the genotoxic effect of 8-MOP is several times higher than its cytotoxicity. Key words: 8-methoxypsoralen, lux biosensor, E. coli, colD gene promoter, SOS response, UV radiation, adducts.

This study investigates the phytochemical composition and biological activities of six solvent extracts-hexane, ethyl acetate, and hydroethanolic fractions-from the roots and aerial parts of Ajania fastigiata (C. Winkl.) Poljakov (Asteraceae), collected in the mountain regions of southeastern Kazakhstan. Phytochemical analysis revealed that the aerial ethyl acetate extract contained the highest levels of bioactive metabolites, including phenolic compounds (34.22 ± 1.98 mg GAE/g), flavonoids (2.74%), organic acids (1.91%), and coumarins (0.54%). Antioxidant potential was evaluated using DPPH and ABTS radical-scavenging assays, with values of SC50 = 133.98 ± 12.91 µg/mL (DPPH) and 209.21 ± 3.70 mg Trolox/g (ABTS). The minimum inhibitory concentration (MIC), determined by the broth microdilution method, showed strong antimicrobial activity of the aerial ethyl acetate fraction, particularly against P.aeruginosa (MIC = 9 µg/mL) and E.faecalis (MIC = 39 µg/mL). Cytotoxicity was assessed by the MTT assay on NIH-3T3, K562, and Saos cell lines. The aerial ethyl acetate fraction exhibited selective cytotoxic effects, with IC50 values of 210 µg/mL for K562 and 360 µg/mL for Saos, while showing negligible activity toward normal fibroblasts (IC50 > 2500 µg/mL). These results confirm the pharmacological potential of A.fastigiata as a promising natural source of antioxidant, antimicrobial, and anticancer agents.

Abstract As a result of this study, the first representative of coumarin-3-carboxylic acid containing a furopyridine fragment was obtained. It was demonstrated that this coumarin-3-carboxylic acid readily forms salts with various organic amines, diamines, as well as with alkali metal cations (Na, K) and ammonia. Testing of all obtained compounds against tumor cell lines M-HeLa and HuTu 80 revealed that a number of salt structures (specifically those based on 2-aminopyridine, 2,6-diaminopyridine, or sodium) exhibit cytotoxicity comparable to the reference drug, 5-fluorouracil.

A new iodine-dextrin-lithium complex (IDLC) was synthesized and structurally characterized as a hybrid supramolecular system combining antiseptic, stabilizing, and biocompatible components. The compound integrates iodine as the primary antimicrobial agent, lithium as a coordination and stabilization element, and dextrin as a biodegradable polysaccharide matrix enabling sustained release. Physicochemical analyses confirmed the formation of a uniform, thermally stable complex. Biological evaluation revealed strong bactericidal activity, with minimum bactericidal concentrations (MBC) ranging from 1.95 to 15.63 µg ml-1 against both Gram-positive and Gram-negative pathogens, including multidrug-resistant Staphylococcus aureus and Acinetobacter baumannii. The complex also demonstrated inhibitory effects in virus-infected MDCK cell cultures. Cytotoxicity studies showed CC50 = 0.23-0.48 mg ml-1 on human MNCs and more than 10 mg ml-1 on MDCK cells, confirming low toxicity. The results of the study on the cytotoxicity of IDLC suggest a cytotoxic effect on tumor cell lines such as HepG2, HeLa, AGS, K562, and H9, as well as normal cell lines MeT-5A. Therefore, it can be estimated that IDLC has the potential to be a new drug with antitumor activity against these cell lines. The research confirms that iodine can be effectively stabilized within a dextrin-lithium framework to yield a biologically active, thermally resistant complex, suitable for pharmaceutical use.

A strategy to create highly effective antimicrobial agents was proposed based on the conjugation of three functional components: a cationic quaternary ammonium salt (QAS) that exerts a membrane-disrupting effect and promotes selective accumulation on bacterial surfaces; a phosphonamidate linker for controlled activation; and a sterically hindered phenol (SHP) fragment as a potential redox component. This approach enabled the preparation of 40 target phosphonamidate–SHP/QAS hybrids in high yields (88–98%). Evaluation of their antimicrobial activity against major pathogens and methicillin-resistant Staphylococcus aureus (MRSA) revealed high potency against Gram-positive bacteria. The lead compounds achieved minimum inhibitory concentration (MIC) values of 0.7–2.8 μM, which is up to 10 times lower than that of the reference drug, norfloxacin. Mechanistic studies confirmed that these hybrids disrupt the bacterial membrane. In addition, an increase in intracellular ROS levels was observed for the most active compound. The SHP/QAS hybrids retained high activity against S. aureus ATCC 209P after 17 passages and showed low cytotoxicity (SI = 62–92) and negligible hemolysis. These properties indicate that this approach may offer a useful strategy for developing antibacterial agents with a potentially lower risk of inducing conventional resistance mechanisms.

Due to the increasing threat of antibiotic resistance and the emergence of new pathogenic strains, the development of effective combined therapeutic agents represents a crucial direction in the fight against infections. Within this study, several compounds were synthesized in which iodine is present in a coordination complex with antibiotics—sodium sulfadimidine and gentamicin sulfate. The physicochemical parameters of these compounds were investigated using capillary electrophoresis and UV spectroscopy, along with their cytotoxicity, antimicrobial, and antiviral activities. As a result of this work, two stable compounds, KC-246 and KC-248, were synthesized, demonstrating virus-inhibitory activity against herpes simplex virus and influenza A under extremely low cytotoxicity levels of 0.018–0.106 mg/mL. Additionally, they exhibited antimicrobial activity against representatives of the families Staphylococcaceae, Pseudomonadaceae, Enterobacteriaceae, Enterococcaceae, and yeast-like fungi. The minimum bactericidal concentrations (MBCs) ranged from 0.794 µg/mL to 0.198 µg/mL (KC-246) and from 2.093 µg/mL to 0.523 µg/mL (KC-248).

Background: Iodine-based antimicrobial systems remain highly attractive due to their broad-spectrum activity; however, the clinical application of free iodine is limited by its instability and cytotoxicity. This study aimed to develop polyethylene glycol (PEG)-based hydrophilic ointment formulations containing a dextrin/polyvinyl alcohol/iodine complex (D/PVA/I) and to evaluate their physicochemical properties, antimicrobial activity, and cytotoxicity. Methods: Hydrophilic ointment formulations containing 2.5%, 5.0%, and 10.0% D/PVA/I were prepared using a PEG-based matrix composed of PEG 4000, PEG 400, and glycerol. Physicochemical characterization included organoleptic evaluation, pH measurement, rheological analysis, and UV–visible (Ultraviolet–visible) spectroscopy. Antimicrobial activity was assessed using agar diffusion and minimum bactericidal concentration (MBC) assays against Staphylococcus aureus, Escherichia coli, Enterococcus hirae, and Pseudomonas aeruginosa. Cytotoxicity was evaluated in Madin–Darby Canine Kidney (MDCK) cells using the MTT assay. Results: All formulations exhibited homogeneous semisolid structure and physiologically acceptable pH values (4.94–5.45). Rheological analysis demonstrated non-Newtonian pseudoplastic (shear-thinning) behavior. The flow behavior index (n) ranged from 0.03 to 0.33 according to the Ostwald–de Waele model, confirming shear-thinning characteristics, while viscosity increased with increasing D/PVA/I concentration. UV–visible spectroscopy confirmed the presence of triiodide ions (I3−), characterized by absorption maxima at approximately 287 and 350 nm, indicating preservation of active iodine species within the PEG matrix, while placebo (blank) formulation analysis confirmed the absence of corresponding absorption bands, demonstrating that the PEG-based matrix does not contribute to the characteristic spectral features. The formulations demonstrated broad-spectrum antimicrobial activity, with MBC values ranging from 0.01 to 0.02 µg/mL. Cytotoxicity studies revealed moderate toxicity of the D/PVA/I complex (CC50 = 0.82%) (50% cytotoxic concentration (CC50) and significantly lower toxicity of the PEG-based ointment base (CC50 = 18.38%). Conclusions: The developed PEG-based hydrophilic ointment formulations containing the D/PVA/I complex demonstrated favorable physicochemical characteristics, stability of iodine species, pronounced antimicrobial activity, and acceptable cytotoxicity profiles. These findings highlight the potential for the developed systems to be promising topical antimicrobial formulations.

Iodine-based antimicrobial materials are widely considered for biomedical applications due to their broad-spectrum antimicrobial activity; however, their potential genotoxicity requires systematic evaluation prior to clinical use. The aim of this study was to assess the genotoxic potential of a dextrin–polyvinyl alcohol–iodine (D/PVA/I-1) complex in accordance with OECD guidelines. Genotoxicity was evaluated in vitro using two complementary assays: the mammalian cell micronucleus test (OECD TG 487) in L5178Y TK+/− cells and the bacterial reverse mutation assay (Ames test, OECD TG 471) using Salmonella typhimurium strains TA98, TA100, TA1535, TA1537 and Escherichia coli WP2 uvrA, both in the presence and absence of metabolic activation (S9). In the micronucleus assay, no statistically significant increase in micronucleus frequency was observed at concentrations ranging from 0.039 to 0.625 mg/mL compared with the negative control. In the Ames test, no increase in revertant colony numbers was detected in any tested bacterial strain at concentrations up to 1250 μg/plate. At higher concentrations, pronounced cytotoxic and bactericidal effects were observed without evidence of mutagenic activity. Overall, the results demonstrate that the D/PVA/I-1 complex does not exhibit genotoxic or mutagenic potential under the tested in vitro conditions, indicating a favorable genetic safety profile and supporting further preclinical evaluation for potential biomedical applications.

Rice husk (RH) is a renewable siliceous lignocellulosic waste providing a unique, greener and less toxic alternative to conventional synthetic polymeric precursors in the production of carbon-based materials for biomedical applications. In this work we studied the porous structure of RH-lignin-based activated carbon produced in the form of honeycomb carbon monoliths and assessed their potential as hemoadsorbents for blood purification in the treatment of patients with serious medical conditions, such as kidney failure and sepsis. To determine their clinical suitability for such an application, the hemocompatibility and cytotoxicity of the monoliths were investigated using the standard ISO guidelines. The monoliths did not cause any changes in the cell viability or cell lysis. High micro/mesoporosity and surface chemistry of the initial monolith-C, N- and P-doped nanostructured carbon honeycomb monoliths were established by low-temperature nitrogen adsorption (LTNA) studies, mercury porosimetry data (MIP), SEM/EDS analysis and FT-IR spectroscopy. The micro-mesoporous, activated carbon-based filtration/adsorbent prototype devices, in the form of three-dimensional (3D) carbon matrix, functionalized with ion-exchange amino- and phosphate groups and encased in polyolefin heat shrink cable sleeve, have been developed with the capacity to remove protein-bound uremic toxins (PBUTs), such us PCS and IS, as well as inflammatory cytokines (IL-6 and IL-8) from human plasma in a flowing model system. The ammoxidized monolith-N, derived from the monolith-C, had the highest removal efficiency (40.05% for PCS, and 28.4% for IL-6). By contrast, phosphorylated monolith-P demonstrated the highest removal efficiency (54.62% for IS, and 54.4% for IL-8), whilst the monolith-C has the lowest removal efficiency for these adsorbates. These results do not correlate with the LTNA and MIP study results, suggesting that the interaction of surface chemical functional groups with the solutes play key roles in the adsorption mechanism. The ion-exchange mechanism of PBUTs and inflammatory cytokine chemisorption by the monoliths, modified with surface N- and P-containing functional groups, has been proposed.
